Essential safety protocols for handling research peptides: PPE, waste disposal and contamination prevention.
Biosafety in the Handling of Research Peptides
Handling lyophilized synthetic peptides requires biosafety protocols BSL-1 / BSL-2 depending on context. Specific risks: contamination of the compound, exposure to fine powders, handling of sharps and disposal of waste.
Applicable levels
| Nivel | Cuándo aplica |
|---|---|
| BSL-1 | Peptides puros no infecciosos en bench abierto |
| BSL-2 | Combinación con cultivos celulares, sangre o muestras biológicas |
Basic PPE
- Closed lab coat, nitrile gloves, safety glasses.
- N95 mask when handling bulk lyophilized powders.
- Hair tied back, closed-toe footwear.
Workspace
- Non-porous surface easy to decontaminate.
- Illumination ≥500 lux.
- Dedicated sink.
- Biosafety cabinet class II for sensitive powders or work with live cells.
Reconstitution protocol
- Lavar manos y colocar EPP.
- Descontaminar superficie con isopropanol 70%.
- Remove the vial and allow it to equilibrate to room temperature (~10 min).
- Clean the stopper with isopropanol.
- Inyectar diluyente deslizando contra la pared.
- Sin agitación vigorosa: rotar suavemente.
- Etiquetar con fecha, diluyente, concentración.
- Conservar refrigerado en empaque profesional original.
Sharps handling
- Approved rigid container.
- Never recap needles with both hands.
- Set up needle + syringe as a single unit.
- Change the container at 75% capacity.
Decontamination
| Superficie | Agente | Tiempo |
|---|---|---|
| Bench | Isopropanol 70% | 30 s |
| Salpicaduras | Etanol 70% + agua | Inmediato |
| Material reutilizable | Autoclave 121 °C | 20 min |
Waste management
- Empty vials: decontaminate with ethanol → chemical waste.
- Syringes: sharps container → biohazardous waste route.
- Diluents: drain disposal only if local regulations permit.
Common errors
- Reconstituir varios viales sin etiquetar (contaminación cruzada).
- Vial sacado del refrigerador inmediatamente (condensación interna).
- Reutilizar jeringas o agujas.
- Conservar reconstituidos en frascos no estériles.
- No documentar fecha de reconstitución.
Mandatory documentation
- Logbook with batch, date, diluent, concentration, operator.
- COA record filed by batch.
- Incident log.
Recommended training
- Institutional BSL-1 course.
- Sharps handling.
- Biological spills.
- First aid for needle exposure.
