IGF-1 LR3: extended half-life analog of insulin-like growth factor. Mechanism of action, comparison with MGF, PEG MGF y ACE-031.
What is IGF-1 LR3?
IGF-1 LR3 (Long R3 Insulin-like Growth Factor-1) is a synthetic analog of insulin-like growth factor 1 (IGF-1) with two key modifications that confer pharmacological properties superior to native IGF-1.
Structural Modifications
- Arg3 substitution: The glutamine at position 3 is replaced by arginine
- N-terminal extension: Addition of 13 amino acids at the N-terminal end
- Result: 83 amino acids vs. 70 of native IGF-1
- Molecular weight: ~9,111 Da
Functional Consequences
These modifications produce:
- Drastic reduction in affinity for IGFBPs (proteínas de unión a IGF): ~1000x menor afinidad
- Greater bioavailability: By not being sequestered by IGFBPs, more free IGF-1 LR3 is available to activate receptors
- Extended half-life: 20-30 horas vs. ~15 minutos del IGF-1 nativo
- Greater in vivo potency: 2-3x more potent than IGF-1 in proliferation assays
Mechanism of Action
Signaling via IGF-1R
IGF-1 LR3 activates the IGF-1 receptor (IGF-1R), a receptor tyrosine kinase:
- Binding to IGF-1R: Dimerización y autofosforilación del receptor
- Activation of IRS-1/2: Fosforilación de sustratos del receptor de insulina
- PI3K/Akt pathway: Señalización de supervivencia celular y síntesis proteica
- Ras/MAPK/ERK pathway: Proliferación celular y diferenciación
- mTOR: Activation of the protein synthesis machinery

The GH/IGF-1 Axis
To understand IGF-1 LR3, it is necessary to contextualize the somatotropic axis:
- Hypothalamus → GHRH (estimulador) + Somatostatina (inhibidor)
- Anterior pituitary → Secreción de GH
- Liver → Producción de IGF-1 (endocrino)
- Local tissues → IGF-1 production (autocrine/paracrine)
IGF-1 mediates approximately 70% of the anabolic effects attributed to GH.
Comparison with Related Growth Factors
MGF (Mechano Growth Factor)
MGF is a splice variant of IGF-1 (IGF-1Ec in humans):
- Origin: Expressed locally in muscle in response to mechanical stress
- Function: Activation of muscle satellite cells
- Mechanism: Signaling different from canonical IGF-1
- Half-life: Very short (~minutes) due to rapid degradation
- Action: Predominantly autocrine/paracrine
PEG MGF (PEGylated MGF)
Pegylated version of MGF for greater stability:
- Modification: Conjugated polyethylene glycol
- Half-life: Significantly greater than native MGF
- Distribution: Systemic vs. local of MGF
- Application: Muscle regeneration research
ACE-031 (Myostatin Decoy Receptor)
ACE-031 is not a growth factor but an inhibitor of myostatin signaling:
- Structure: ActRIIB-Fc fusion protein
- Mechanism: Sequesters myostatin, activin, and GDF-11
- Effect: Muscle hypertrophy via disinhibition
- Complementarity: Acts via a different pathway than IGF-1

IGFBPs: Modulators of Activity
IGF-binding proteins (IGFBP-1 to IGFBP-6) regulate the bioavailability of IGF-1:
- IGFBP-3: Main plasma carrier (~75% of circulating IGF-1)
- IGFBP-5: Enhancer of IGF-1 activity in bone
- IGFBP-1: Regulated by insulin, it modulates postprandial IGF-1 activity
IGF-1 LR3 evades this regulatory system, which explains its greater potency but also the need for careful dosing in research.
Research Applications
Muscle Biology
- Studies of muscle hypertrophy and regeneration
- Activation of satellite cells
- Transition of type IIb → type IIa fibers
Neuroscience
- Neuroprotection and neurogenesis
- CNS myelination
- Research in neurodegenerative diseases
Metabolism
- Nutrient partitioning
- Insulin sensitivity
- Glucose metabolism
Technical Considerations
- Reconstitution: 0.1M acetic acid or bacteriostatic water
- Storage: -20°C (lyophilized), 2-8°C (reconstituted, max 30 days)
- Sensitivity: Avoid freeze-thaw cycles
- Incompatibilities: Avoid neutral/basic pH during storage
Conclusion
IGF-1 LR3 is the most potent and bioavailable form of IGF-1 available for research, thanks to its modifications that evade the regulatory system of IGFBPs. Together with MGF, PEG MGF and ACE-031, it forms an arsenal of tools for research on the regulation of tissue growth and protein synthesis.
